Details
This handy kit is for small-scale trial expression tests using plasmid DNA templates. It contains transcription premix and translation reagents to perform eight 226-µL reactions of wheat germ protein synthesis. It also contains the pEU-E01-MCS vector specifically designed for the wheat germ cell-free system, into which genes of interest are inserted to produce plasmid DNA templates. The pEU-E01-DHFR vector contained in the kit is used as positive control and expected to yield about 12 µg of DHFR protein in a 226-µL reaction.
Reaction steps
- Plasmid DNA construction
- Preparation of plasmid DNA for transcription
- Transcription using plasmid DNA template, incubation at 37 ℃ for 6 hours
- Translation reagents preparation
- Translation of target protein by bi-layer reaction, incubation at 15 ℃ for 12-20 hours
